What information do I need to provide to assess sequence pricing and turnaround time?

We require the DNA or amino acid sequence you need synthesized. If codon optimization is required, specify the host species. Additionally, indicate any required restriction sites at the 5' and/or 3' ends, any internal modifications to avoid, and if you need additional sequences added (e.g., Kozak sequence, stop codons, etc.).Or you can download the "Tsingke Gene Synthesis Order Form" from our website and email it to info@tsingke.com.cn. What contaminants are present in the Crude Product?

Crude oligonucleotides contain the desired product, failure sequences and chemically modified by-products.

Do Tsingke’s oligos come with 5' phosphorylation

Synthesized oligos have a hydroxyl group at the 5' end and are not phosphorylated. 

If 5' phosphorylation is required, you can phosphorylate them using polynucleotide kinase, or request phosphorylation at the 5' or 3' end when palce your order.

What is the maximum length Tsingke can synthesize oligos?

We specialize in synthesizing RNA oligos up to 180 nucleotides and DNA oligos exceeding 250 nucleotides.

How to Store Oligos?

Oligo primers in freeze-dried state are highly stable and can be stored at -20°C for at least one year. After reconstitution, dissolved oligos can be stored as a 100 μmol/L stock solution, aliquoted into multiple tubes, and kept at -20°C for at least six months (frequent freeze-thaw cycles may reduce shelf life). Before use, dilute the concentrated solution to the working concentration for your experiments.

Each OD of primer needs to be diluted with water to a concentration of 100 μmol/L (equivalent to 100 pmol/μL).You could add an appropriate amount of nuclease-free double-distilled water (pH >6.0) or TE buffer (pH 7.5-8.0) according to your experimental needs.

Before opening the vial, centrifuge it at 3000-4000 rpm for 1 minute to prevent the loss of primers when opening.
