These lipids were mixed at a molar ratio of 50/10/38.5/1.5 in ethanol to form the lipid phase. 2'-O-methyl-modified siRNA was synthesized by Beijing Tsingke Biotechnology Co., Ltd. and dissolved in 25 mM acetate buffer (pH = 4.0) to form the aqueous phase. The two phases were mixed using a Microfluidic chip (Microfluidic ChipShop, Germany) at a volume flow ratio of 3:1 (water/ethanol).
