Dual-Luciferase Reporter Assay To construct a luciferase reporter gene vector containing the MeCP2 promoter, the full-length MeCP2 promoter, containing wild-type (WT) and mutant (Mut), were respectively cloned into pGL3-basic vectors (Qingke Biotechnology, Beijing, China), thereby generating the luciferase reporter vectors. Then HEK-293T cells were co-transfected with the corresponding plasmids with or without C/EBP-β vector (Qingke Biotechnology, Beijing, China), using the jetPRIME® transfection reagent (Polyplus Transferion® SA, Strasbourg, France). After 48 h, the Dual Luciferase Reporter Gene Assay Kit (Biosharp, Hefei, Anhui, China) was used to detect the firefly luciferase and Renilla luciferase activities.
