Lnc237 regulates duck myoblasts proliferation and differentiation via the miR-22-5p/MYL9 axis

Abstract

The full length sequence of lnc237 was shown in the Table S1. The overexpression vector PCD-lnc237 (pcDNA3.1(+) + lnc237) was synthesized by Tsingke Biotechnology (Beijing, China), including the vectors psiCHECK2-lnc237-MUT, psiCHECK2-MYL9-WT, psiCHECK2-MYL9-MUT and the small interfering RNA (si-RNA) targeting lnc237 and MYL9 (Table S2). Then, dual luciferase expression vectors psiCHECK2-lnc237-WT containing miR-22-5p binding sites and the overexpression vector PCD-MYL9 (pcDNA3.1(+) + MYL9) were constructed. The miR-22-5p mimics and inhibitors were synthesized by Sangon Biotech (Shanghai, China). The primers used in this study were all synthesized by Tsingke Biotechnology, and the sequence information was shown in the Table S2. All vectors were verified by sequencing, and all vectors, mimics, and inhibitors were transfected into cells with Lipo8000TM (Beyotime, Shanghai, China) according to the instructions of the manufacturer.
