CRISPR/Cas9-mediated knockout of CRE To knock out one IGF2BP2-linked CRE (located at chr3:185995289-185995789 region) from the genome, lentivirus (IGF2BP2-CRE4-C4 KO) containing single-guide RNA (sgRNA) targeting specific CRE region and Cas9 was designed and synthesised by Tsingke. The sgRNA designed based on the sequence of CRE region obtained from NCBI were cloned into lentiCRISPR v2-cmv-ZsGreen-puro vector and vehicle vector (IGF2BP2-CRE4-C4 WT) was used as negative control.
