Primers for the goat ACOT7 gene were designed based on the ACOT7 gene (accession number: NC_030823.1) and the respective mRNA sequence (accession number: XM_013970256.2) retrieved from the NCBI GenBank database. Exon-targeted primers were designed using Primer Premier 5.0 software (PREMIER Biosoft International, Palo Alto, CA, USA) and synthesized by Qingke Biotechnology Co., Ltd., Chongqing, China. The detailed information regarding the primers is shown in Table 9.|||The PCR program was set as follows: predenaturation at 94 °C for 5 min; denaturation at 94 °C for 30 s, annealing at 60 °C for 30 s, extension at 72 °C for 30 s, for a total of 35 cycles; final extension at 72 °C for 10 min; and storage at 4 °C. The amplified PCR products were examined using 1.0% agarose gel electrophoresis (Fig. 8), and qualified samples were sent for sequencing at Beijing Qingke Biotechnology Co., Ltd., China.
